Evaluation of ten (10) SARS-CoV-2 rapid serological tests in comparison with WANTAI SARS-CoV-2 ab ELISA in Burkina Faso, West Africa

Henri Gautier Ouedraogo, Abdou Azaque Zoure, Tegwinde Rebeca Compaoré, Herve Ky, Sylvie Zida, Dezemon Zingué, Oumarou Ouedraogo, Serge Théophile Soubeiga, Tani Sagna, Charlemagne Dabiré, Dinanibè Kambiré, Dramane Zongo, Albert Théophane Yonli, Abdoul Rahamani Nikiema, Désiré Nezien, Gnintassa Cyrille Bansé, Brice Wilfried Bicaba, Sophie Perier, Charles Sawadogo, Zakariya Yabre, Lassana Sangare

BackgroundThe aim of this study was to evaluate the performance of ten (10) SARS-CoV-2 serological rapid diagnostic tests in comparison with the WANTAI SARS-CoV-2 Ab ELISA test in a laboratory setting.Materials and methodsTen (10) SARS-CoV-2 serological rapid diagnostic tests (RDTs) for SARS-CoV-2 IgG/IgM were evaluated with two (2) groups of plasma tested positive for one and negative for the other with the WANTAI SARS-CoV-2 Ab ELISA. The diagnostic performance of the SARS-CoV-2 serological RDTs and their agreement with the reference test were calculated with their 95% confidence intervals.ResultsThe sensitivity of serological RDTs ranged from 27.39 to 61.67% and the specificity from 93.33 to 100% compared to WANTAI SARS-CoV-2 Ab ELISA test. Of all the tests, two tests (STANDARD Q COVID-19 IgM/IgG Combo SD BIOSENSOR and COVID-19 IgG/IgM Rapid Test (Zhejiang Orient Gene Biotech Co., Ltd)) had a sensitivity greater than 50%. In addition, all ten tests had specificity greater than or equal to 93.33% each. The concordance between RDTs and WANTAI SARS-CoV-2 Ab ELISA test ranged from 0.25 to 0.61.ConclusionThe SARS-CoV-2 serological RDTs evaluated show low and variable sensitivities compared to the WANTAI SARS-CoV-2 Ab ELISA test, with however a good specificity. These finding may have implications for the interpretation and comparison of COVID-19 seroprevalence studies depending on the type of test used.

Evaluation of the Analytical Performance of the STANDARD Q COVID‐19 Antigenic Test in Suspected COVID‐19 Cases in Bobo‐Dioulasso, Burkina Faso

BackgroundThe use of alternative technique such as rapid antigen diagnostic tests (Ag‐RDTs) is needed in countries with limited resources for fast tracking of COVID‐19 cases. This study evaluated the performance of the STANDARD Q COVID‐19 antigen test compared with reverse transcriptase polymerase chain reaction (RT‐PCR) among suspected COVID‐19 cases using nasopharyngeal and/or oropharyngeal swabs in Bobo‐Dioulasso, Burkina Faso.MethodsA cross‐sectional study was conducted from April to September 2021. The respiratory secretions of suspected COVID‐19 cases were collected at the Virology Laboratory of Sourô Sanou University Hospital in Bobo‐Dioulasso. The respiratory secretions consisted of nasopharyngeal or combined nasopharyngeal/oropharyngeal swabs. Each sample was analyzed using the STANDARD Q COVID‐19 Ag test and RT‐PCR on the Abbott m2000 RealTime system.ResultsA total of 152 respiratory specimens were collected from suspected COVID‐19 cases and processed in this study. The sensitivity and specificity of the Ag‐RDT were 26.32% (9.15–57.20) and 98.65% (92.7–99.9), respectively, in the suspected cases with nasopharyngeal swabs from suspected cases, while a sensitivity of 25.0% (3.2–65.1) and a specificity of 98.65% (92.7–99.9) were shown with combined nasopharyngeal/oropharyngeal swabs from suspected cases. The accuracy and Cohen′s kappa of the test were 83.87% and 0.36 in suspected cases with nasopharyngeal swabs from suspected cases, while they were 83.37% and 0.32 in oropharyngeal swabs. In our study, Ag‐RDT was not sensitive on the specimens of suspected COVID‐19 cases with Ct values ≥ 20, whereas its sensitivity increased to 41.18% (18.4–67.1) on specimens with Ct < 20.ConclusionThe study findings have shown that the Ag‐RDT is useful in suspected COVID‐19 cases. However, the sensitivity of the STANDARD Q COVID‐19 Ag test was low with minimal agreement in comparison to RT‐PCR on the Abbott m2000rt system.

Evaluation of the community response to HIV and AIDS: Learning from a portfolio approach

Rosalía Rodriguez-García, David Wilson, Nick York, Corinne Low, N'Della N'Jie, Rene Bonnel

While communities have played a large role in the HIV/AIDS response, their contributions and innovative approaches to HIV prevention, treatment, care and support have not always been the focus of systematic and rigorous evaluations. To address this gap, the World Bank led an evaluation of the impact of the community response to HIV, including country studies in Burkina Faso, India, Kenya, Lesotho, Nigeria, Senegal, South Africa and Zimbabwe over a three-year period. Due to the complexity and varied nature of community responses, the evaluation attempted to determine the results that investments have produced at the community level by applying a mixed method approach: Randomized Controlled Trials, quasi-experimental studies, qualitative studies and analytical studies including financial data. Specifically, the studies examined a typology of community response and the flow of funds to community-based organizations, while investigating the impact of the community responses on (1) knowledge and behavior, (2) use of services, (3) social transformation, and (4) HIV incidence. This editorial summarizes the results of this evaluation portfolio, finding that investments in communities have produced significant results, including, improved knowledge and behavior, and increased use of health services, and even decreased HIV incidence. Evidence on social transformation was more mixed, with community groups found to be effective only in some settings. Each study in the evaluation provides a partial view of how communities shape the local response; however, taken together they corroborate the common wisdom that communities can be a vital part of the global HIV/AIDS response.

Evaluation of the Diagnostic Performances of the SD-Bioline®HBeAg Rapid Test Used Routinely for the Management of HBV-Infected Individuals in Burkina Faso

Hepatitis B e antigen (HBeAg) is a marker of wild-type hepatitis B virus replication. In resource-limited countries where access to enzyme-linked immunosorbent assay (ELISA) remains a challenge, rapid diagnostic tests (RDT) constitute a good alternative. The HBeAg status is employed to evaluate eligibility for antiviral therapy and to prevent the transmission of hepatitis B from mother to child (PMTCT). The objective of this study was to assess the diagnostic performance of the SD-Bioline®HBeAg RDT commonly used for detecting HBeAg in laboratories in Burkina Faso. The sample panel used was collected from HBsAg-positive patients received in the laboratory for the detection of HBeAg with the rapid test. The samples were retested for HBeAg using the VIDAS HBe/Anti-HBe enzyme-linked fluorescent assay (ELFA) (Gold standard). Then, the viral load (VL) of HBV DNA was determined using the GENERIC HBV CHARGE VIRLAE kit (GHBV-CV). The diagnostic performances of the SD-Bioline®HBeAg and its agreement with the gold standard were calculated with their 95% confidence intervals. Overall, 340 sera obtained from HBsAg-positive patients were included in this evaluation Compared to the VIDAS HBe/Anti-HBe ELFA test, the sensitivity (Se) and specificity (Sp) of the SD-Bioline®HBeAg test were 33.3% and 97.9%, respectively. The concordance between the two tests was 0.42. Depending on the viral load, the Se and Sp varied from 8.8% and 98.3% for a VL < 2000 IU/mL to 35.5% and 98.4% for a VL > 2,000,000 IU/mL. The results showed a low sensibility of the SD-Bioline®HBeAg RDT test, indicating that its use is inappropriate for the clinical management of HBV-infected patients. They also highlight the urgent need to develop HBeAg rapid tests with better sensitivities.

Evaluation of the enhanced meningitis surveillance system, Yendi municipality, northern Ghana, 2010–2015

Basil Benduri Kaburi, Chrysantus Kubio, Ernest Kenu, Kofi Mensah Nyarko, Jacob Yakubu Mahama, Samuel Oko Sackey, Edwin Andrew Afari

BackgroundMeningitis is the inflammation of the meninges of the brain and or spinal cord. Global mortality rates vary from 2% to 30%. Epidemic meningitis remains a public health concern along the meningitis belt of Africa. Despite the operation of an enhanced meningitis surveillance system in Ghana, institutional mortality rates are estimated to range from 36% to 50%. In 2014, Yendi recorded 83 confirmed cases; with focal epidemics in some sub-municipals. We evaluated the system over a five-year period to find out whether it was achieving its objectives of systematic collection and analyses of data for the prevention or early detection of meningitis epidemics.MethodsWe used cross-sectional design. Both qualitative and quantitative data from Yendi Municipality between January 2010 and December 2015 were collected and analyzed. The updated guidelines for evaluating surveillance systems from Centers for Disease Control and Prevention were used. Content analysis was performed on the responses of key informants. Surveillance data was analyzed using MS-Excel.ResultsFifteen healthcare workers were interviewed. For the period under evaluation, the annual incidence of meningitis ranged from 1.6/100,000 in 2012 to 62.6/100,000 in 2014. The average case fatality rate for the period was 8.3%. The system was sensitive, representative, and acceptable. The predictive value positive was 100% from 2010 to 2014 and 63.3% in 2015. Data quality was good, but timeliness of reporting was poor.ConclusionsThe enhanced meningitis surveillance system in Yendi Municipality is achieving most of its objectives. However, financial constraints and poor personnel motivation pose threats to the sustainability of the system.Electronic supplementary materialThe online version of this article (doi:10.1186/s12879-017-2410-0) contains supplementary material, which is available to authorized users.

BackgroundAn issue of particular concern is the impact of the 2019 novel coronavirus (2019 nCOV) on the people coinfected with the Human Immuno-deficiency Virus (HIV) and/or tuberculosis (TB). Unfortunately, this interaction has not been well explored in African despite the large proportion of these risk populations living with HIV and/or patients and/or tuberculosis (TB) in the African region. This study aims to design a research protocol for assessment of the impact of coronavirus disease 2019 (COVID-19) on these risk populations in response to COVID-19 strategic plans in Burkina Faso by generating serological, epidemiological, virological, clinical and socio-anthropological evidence-based data.MethodsA multidisciplinary research will be conducted in the city of Bobo-Dioulasso, Burkina Faso using mixed methods. Data will be collected from a cohort of people living with HIV and/or TB patients in the city (i) to determine the proportion of people with specific antibodies against the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) using retrospective data ; (ii) to determine the proportion of people infected with Covid-19 and the dynamics of viral loads and antibodies in these people based on prospective data; (iii) to identify circulating SARS-COV-2 variants and novel biomarkers using prospective data ; (iv) to analyze perceptions, community experiences and response strategies during the public health emergencies imposed by COVID-19 through a qualitative study.DiscussionThis study will generate factual and comprehensive data that will contribute in improving response strategies to COVID-19 and the other possible emerging diseases with keen interest on the risk populations living with HIV and/or TB infected patients.Supplementary InformationThe online version contains supplementary material available at 10.1186/s12879-023-08076-4.

Evaluation of the implementation fidelity of the seasonal malaria chemoprevention intervention in Kaya health district, Burkina Faso

BackgroundBurkina Faso implemented the seasonal malaria chemoprevention (SMC) in 2014 in seven pilot health districts, following the new recommendation by the WHO in 2012 for the prevention of the disease in children under five years old, for areas of highly seasonal malaria transmission. The objective of this study was to assess the implementation fidelity of the seasonal malaria chemoprevention strategy in one of the districts, Kaya Health District.MethodologyWe conducted a case study, with a quantitative and qualitative mixed methods. Data were collected after two campaigns of implementation of the intervention, in 2014 and 2015, through a review of specific documents of SMC intervention, and individual interview with key informants (n = 21) involved at various levels in the implementation of the strategy and a household survey with the parents (n = 284) of eligible children for the SMC strategy in 2015 in the Kaya health district. The analysis framework focused on the fidelity of the intervention’s content, its coverage, and its schedule, as well as the potential moderating factors, using the model proposed by Hasson, originally from Carroll.ResultsAll components of the intervention were implemented. Villages and sectors were covered at 100%. In terms of intervention doses received, less than one-third of eligible children (32.3%) received the recommended four doses in 2015. Implementation of the strategy faced some difficulties due to insufficient training of community distributors, inadequate supply of inputs and insufficient financial resources for remuneration, advocacy and supervision, but also because of the contextual constraints due to the rainy season. Moreover, an interaction between the different moderating factors, influencing the degree of implementation of the strategy was noted.ConclusionTaking into account the moderating factors of the implementation is necessary for achieving the highest possible degree of implementation fidelity and then, reach

Evaluation of the physicochemical parameters of edible oils sold in the three cities of Burkina Faso

Kabakdé Kaboré, Kiéssoun Konaté, Hemayoro Sama, Roger Dakuyo, Abdoudramane Sanou, David Bazié, Mamounata Diao, Mamoudou Hama Dicko

AbstractThe edible oil needs of African countries are met by imported or locally produced ones. Therefore, consumers are generally confronted with a choice of edible oils of poorly controlled quality. However, quality control of edible oils for local consumption is of high necessity. This study aimed to assess the quality of edible oils sold and consumed in some cities in Burkina Faso. Oil samples collected in the cities of Dédougou, Koudougou, and Nouna were used for several analyses. Oil samples from palm, refined and unrefined cottonseed, and groundnut were collected. Standard methods were used to assess the physicochemical quality parameters of the oils, including the peroxide value, water and volatile matter content, acid value, traces of soap, and mineral oil contents. The parameters varied significantly depending on the oil type, but not by the locality of origin. The peroxide indices had varied from 3.24 to 39.99 mEq O2/kg oil. The acid indices varied from 0.22 mg KOH/g to 1.24 mg KOH/g. The water and volatile matter contents ranged from 0.04% to 0.88%. The test for traces of soap gave values ranging from 0 to 76 ppm. For the mineral oil test, four samples of cottonseed oil collected in Dedougou gave positive results. Compared to international reference standards, in particular the Codex Alimentarius standard, it may constitute a health risk for consumers. The poor storage, distribution, and marketing conditions of the oils could explain their poor quality. In order to provide consumers with quality oils, regular controls must be undertaken in the places where the oils are stored and/or marketed. Oil is an important food for the human organism; however, the oils sold on our markets do not all respect the quality standards. In Burkina Faso, mainly in Koudougou, Dédougou, and Nouna, high levels of mineral oils and traces of soap have been detected, in addition to extensive oxidation, and these oils can therefore constitute a public health problem. Evaluation of the routine implementation of pulse oximeters into integrated management of childhood illness (IMCI) guidelines at primary health care level in West Africa: the AIRE mixed-methods research protocol

Gildas Boris Hedible, Sarah Louart, Désiré Neboua, Laura Catala, Gildas Anago, Abdoul-Guaniyi Sawadogo, G. Désiré Kargougou, Bertrand Meda, Jacques Séraphin Kolié, Adama Hema, Sory Keita, Mactar Niome, Abdoul Salam Savadogo, Lucie Peters-Bokol, Honorat Agbeci, Zineb Zair, Severin Lenaud, Marine Vignon, Solange Ouedraogo Yugbare, Hannatou Abarry, Abdoul Aziz Diakite, Ibrahima Sory Diallo, Franck Lamontagne, Valérie Briand, Désiré Lucien Dahourou, Anthony Cousien, Valéry Ridde, Valériane Leroy

BackgroundThe AIRE operational project will evaluate the implementation of the routine Pulse Oximeter (PO) use in the integrated management of childhood illness (IMCI) strategy for children under-5 in primary health care centers (PHC) in West Africa. The introduction of PO should promote the accurate identification of hypoxemia (pulse blood oxygen saturation Sp02 < 90%) among all severe IMCI cases (respiratory and non-respiratory) to prompt their effective case management (oxygen, antibiotics and other required treatments) at hospital. We seek to understand how the routine use of PO integrated in IMCI outpatients works (or not), for whom, in what contexts and with what outcomes.MethodsThe AIRE project is being implemented from 03/2020 to 12/2022 in 202 PHCs in four West African countries (Burkina Faso, Guinea, Mali, Niger) including 16 research PHCs (four per country). The research protocol will assess three complementary components using mixed quantitative and qualitative methods: a) context based on repeated cross-sectional surveys: baseline and aggregated monthly data from all PHCs on infrastructure, staffing, accessibility, equipment, PO use, severe cases and care; b) the process across PHCs by assessing acceptability, fidelity, implementation challenges and realistic evaluation, and c) individual outcomes in the research PHCs: all children under-5 attending IMCI clinics, eligible for PO use will be included with parental consent in a cross-sectional study. Among them, severe IMCI cases will be followed in a prospective cohort to assess their health status at 14 days. We will analyze pathways, patterns of care, and costs of care.DiscussionThis research will identify challenges to the systematic implementation of PO in IMCI consultations, such as health workers practices, frequent turnover, quality of care, etc. Further research will be needed to fully address key questions such as the best time to introduce PO into the IMCI process, the best SpO2 threshold for d

Evaluation of treatment patterns, healthcare resource utilization and cost of illness for sickle cell disease in Ghana: a private medical insurance claims database study

Kwaku Marfo, Yvonne Dei-Adomakoh, Catherine Segbefia, Duah Dwomoh, Adeline Edgal, Nancy Ampah, Badarinath Chickballapur Ramachandrachar, Kumaresan Subramanyam, Ashok Natarajan, Olufolake Egbujo, Kenneth I. Ataga

BackgroundSickle cell disease (SCD) is a major public health concern in sub-Saharan Africa, accounting for nearly 75% of the global disease burden. The current analysis evaluated patient characteristics, treatment patterns, healthcare resource utilization (HCRU) and associated costs in patients with SCD based on a Private Medical Insurance Database in Ghana.MethodsThis retrospective longitudinal cohort study was conducted using an e-claims database from Ghana (01 January 2015 to 31 March 2021). Patients were stratified by age (0 month to < 2 years, ≥ 2 years to ˂6 years, ≥ 6 years to < 12 years, ≥ 12 years to < 16 years; ≥16 years), vaso-occlusive crisis (VOC) (< 1, ≥ 1 to < 3, and ≥ 3 per year), and continuous enrolment. Study outcomes related to patient characteristics, comorbidities, treatment pattern, HCRU were evaluated for pre- and post-index period (index period was between July 2015 to March 2020). Descriptive analysis was used to analyse different study variables.ResultsThe study included 2,863 patients (mean age: 20.1 years; Min age: 0; Max age: 83; females 56.1%). Overall, 52.2% (n = 1,495) of SCD patients were ≥ 16 years and 17.0% (n = 486) were in the ≥ 2 to ˂6-years age group. The majority of patients aged ≥ 16 years (62.5%) in the database did not have reported VOC episodes, 35.9% of patients had 1 to 3 VOCs per year and 1.5% had ≥ 3 VOCs per year during the follow-up period. Consultation-based prevalence of SCD was 0.5% [95% confidence interval (CI): 0-1.3%] − 1.4% [CI: 0.6-2.2%]. Malaria, upper respiratory tract infection (URTI) and sepsis were the common complications of SCD. Analgesics were the most frequently prescribed medications followed by anti-infectives, hematinics, and antimalarials. Hydroxyurea, a routine standard of care for SCD was under-utilized. SCD patients had median cost incurred for consultation/hospital services of $11.3 (Interquartile range [IQR] $6.2 - $27.2). For patients with VOC, maximum median cost was incurred for medicati

Evaluation of two lead malaria transmission blocking vaccine candidate antibodies in natural parasite-vector combinations

Anais Bompard, Dari F. Da, Rakiswendé S. Yerbanga, Sumi Biswas, Melissa Kapulu, Teun Bousema, Thierry Lefèvre, Anna Cohuet, Thomas S. Churcher

Transmission blocking vaccines (TBV) which aim to control malaria by inhibiting human-to-mosquito transmission show considerable promise though their utility against naturally circulating parasites remains unknown. The efficacy of two lead candidates targeting Pfs25 and Pfs230 antigens to prevent onwards transmission of naturally occurring parasites to a local mosquito strain is assessed using direct membrane feeding assays and murine antibodies in Burkina Faso. The transmission blocking activity of both candidates depends on the level of parasite exposure (as assessed by the mean number of oocysts in control mosquitoes) and antibody titers. A mathematical framework is devised to allow the efficacy of different candidates to be directly compared and determine the minimal antibody titers required to halt transmission in different settings. The increased efficacy with diminishing parasite exposure indicates that the efficacy of vaccines targeting either Pfs25 or Pfs230 may increase as malaria transmission declines. This has important implications for late-stage candidate selection and assessing how they can support the drive for malaria elimination.

Evaluation of two red cell inclusion staining methods for assessing spleen function among sickle cell disease patients in North-East Nigeria

The loss of splenic function is associated with an increased risk of infection in sickle cell disease (SCD); however, spleen function is rarely documented among SCD patients in Africa, due partly to the non-availability of sophisticated techniques such as scintigraphy. Methods of assessing splenic function which may be achievable in resource-poor settings include counting red blood cells (RBC) containing Howell Jolly Bodies (HJB) and RBC containing silver-staining (argyrophilic) inclusions (AI) using a light microscope. We evaluated the presence of HJB—and AI—containing RBC as markers of splenic dysfunction among SCD patients in Nigeria. We prospectively enrolled children and adults with SCD in steady state attending outpatient clinics at a tertiary hospital in North-East Nigeria. The percentages of HJB—and AI-containing red cells were estimated from peripheral blood smears and compared to normal controls. There were 182 SCD patients and 102 healthy controls. Both AI- and HJB-containing red cells could be easily identified in the participants blood smears. SCD patients had a significantly higher proportion of red cells containing HJB (1.5%; IQR 0.7%–3.1%) compared to controls (0.3%; IQR 0.1%–0.5%) (P <0.0001). The AI red cell counts were also higher among the SCD patients (47.4%; IQR 34.5%–66.0%) than the control group (7.1%; IQR 5.1%–8.7%) (P < 0.0001). The intra-observer reliability for assessment of HJB- (r = 0.92; r2 = 0.86) and AI- containing red cells (r = 0.90; r2 = 0.82) was high. The estimated intra-observer agreement was better with the HJB count method (95% limits of agreement, -4.5% to 4.3%; P = 0.579).We have demonstrated the utility of light microscopy in the assessment of red cells containing—HJB and AI inclusions as indices of splenic dysfunction in Nigerian SCD patients. These methods can be easily applied in the routine evaluation and care of patients with SCD to identify those at high risk of infection and initiate appropriate preventive measures.

Ghizlane Slimani, Hicham Lakbiri, Fatima Zahra Sekkat

Conclusion : les AINS ne semblaient pas aug

Evaluative study of a MOOC on knowledge translation in five French-speaking countries

Over the past few years, the RENARD research team has observed a sharp increase in the need for knowledge translation (KT) training. Given the high demand, it has been nearly impossible for the team to provide training entirely in person, and so a massive open online course (MOOC) was developed. Its aim is to promote the use and appropriation of the KT process by practitioners, decision-makers, and others in the public sphere. The goal of this study was to evaluate the MOOC by collecting users’ opinions, reactions, appropriation, and practice changes. A qualitative research design was used. Data were collected through semi-structured individual interviews (n = 16) with professionals from Canada, France, and three West African countries (Burkina Faso, Mali, and Senegal) who had taken the MOOC. All interviews were subjected to thematic content analysis. The MOOC content was generally appreciated and reused by the respondents. The results revealed one main motive for completing the course: the immediate opportunity to apply their learning in their practice environments. However, most respondents deplored the lack of interaction among learners and expressed the need for coaching with an instructor to deepen the topics covered during the course. The results also revealed connection and accessibility issues linked to the Internet network and unstable access to electricity in West African countries. The study highlights the potential of MOOCs for the acquisition of knowledge and competencies by KT professionals. Several recommendations and avenues of exploration were formulated to optimize and improve future designs of MOOCs on KT.

Jean-Pierre Rossi, Martin Godefroid, Christian Burban, Marguerite Chartois, Xavier Mesmin, Pauline Farigoule, Jean-Claude Streito, Astrid Cruaud, Jean Yves Rasplus

Joseph Larmarange, Abdou Diop, Oulimata Diop, Khady Gueye, Catherine Enel, Souleymane Mboup, Pape Niang Diallo, Ndeye C Toure Kane, Sidy Ba, Amsata Sene, Adama Marra, Abdoulaye S Wade, Annabel Desgrées Du Loû

Hicham Kechna, Omar Ouzzad, Khalid Chkoura, Jaouad Loutid, Moulay Ahmed Hachimi, Sidi Mohamed Hanafi

Francisca Monebenimp, Gilles Kagmeni, David Chelo, Yannick Bilong, Ernest Moukouri

Evidence for Divergent Selection on Immune Genes between the African Malaria Vectors, Anopheles coluzzii and A. gambiae

Yoosook Lee, Lattha Souvannaseng, Travis C. Collier, Bradley J. Main, Laura C. Norris, Abdarahamane Fofana, Sekou F. Traoré, Anthony J. Cornel, Shirley Luckhart, Gregory C. Lanzaro

Simple SummaryA comparison of the genomes of the African malaria vectors, Anopheles gambiae and A. coluzzii, revealed that immune genes are highly diverged. Although these two species frequently co-occur within a single site, they occur in distinct larval habitats. Our results taken in the context of known differences in the larval habitats occupied by these taxa support the hypothesis that observed genetic divergence may be driven by immune response to microbial agents specific to these habitats. Strict within species mating may have subsequently evolved in part to maintain immunocompetence which might be compromised by dysregulation of immune pathways in hybrids. We conclude that the evolution of immune gene divergence among this important group of species may serve as a useful model to explore ecological speciation in general.AbstractDuring their life cycles, microbes infecting mosquitoes encounter components of the mosquito anti-microbial innate immune defenses. Many of these immune responses also mediate susceptibility to malaria parasite infection. In West Africa, the primary malaria vectors are Anopheles coluzzii and A. gambiae sensu stricto, which is subdivided into the Bamako and Savanna sub-taxa. Here, we performed whole genome comparisons of the three taxa as well as genotyping of 333 putatively functional SNPs located in 58 immune signaling genes. Genome data support significantly higher differentiation in immune genes compared with a randomly selected set of non-immune genes among the three taxa (permutation test p < 0.001). Among the 58 genes studied, the majority had one or more segregating mutations (72.9%) that were significantly diverged among the three taxa. Genes detected to be under selection include MAP2K4 and Raf. Despite the genome-wide distribution of immune genes, a high level of linkage disequilibrium (r2 > 0.8) was detected in over 27% of SNP pairs. We discuss the potential role of immune gene divergence as adaptations to the different la
