Abdoul Wahab Alassane Mohamed, Koini Moussa, Souna Badio Seyni, Zirbine Abassa Seyni, Akambi Sanoussi Kasoumou, Konguise Ziberou

Seydou Togo, Ouattara Moussa Abdoulaye, Li Xing, Yena Sadio

Fouad Nya, Abdessamad Abdou, Mehdi Bamous, Younes Moutakiallah, Noureddine Atmani, Aniss Seghrouchni, Mahdi Aithoussa, Abdellatif Boulahya

Amine Tarmiz, Imene Mgarrech, Mehdi Slim, Chokri Kortas, Sofiane Jerbi

Narrative review of methods and findings of recent studies on the carriage of meningococci and other Neisseria species in the African Meningitis Belt

Brian M. Greenwood, Abraham Aseffa, Dominique A. Caugant, Kanny Diallo, Paul A. Kristiansen, Martin C. J. Maiden, James M. Stuart, Caroline L. Trotter

AbstractObjectiveTo review the findings of studies of pharyngeal carriage of Neisseria meningitidis and related species conducted in the African meningitis belt since a previous review published in 2007.MethodsPubMed and Web of Science were searched in July 2018 using the terms ‘meningococcal OR Neisseria meningitidis OR lactamica AND carriage AND Africa’, with the search limited to papers published on or after 1st January 2007. We conducted a narrative review of these publications.ResultsOne hundred and thirteen papers were identified using the search terms described above, 20 of which reported new data from surveys conducted in an African meningitis belt country. These papers described 40 surveys conducted before the introduction of the group A meningococcal conjugate vaccine (MenAfriVacR) during which 66 707 pharyngeal swabs were obtained. Carriage prevalence of N. meningitidis varied substantially by time and place, ranging from <1% to 24%. The mean pharyngeal carriage prevalence of N. meningitidis across all surveys was 4.5% [95% CI: 3.4%, 6.8%] and that of capsulated N. meningitidis was 2.8% [95% CI: 1.9%; 5.2%]. A study of households provided strong evidence for meningococcal transmission within and outside households. The introduction of MenAfriVac® led to marked reductions in carriage of the serogroup A meningococcus in Burkina Faso and Chad.ConclusionsRecent studies employing standardised methods confirm the findings of older studies that carriage of N. meningitidis in the African meningitis belt is highly variable over time and place, but generally occurs with a lower prevalence and shorter duration than reported from industrialised countries.

Nasopharyngeal carriage and risk factors of major meningitis pathogens among asymptomatic healthcare workers in paediatric units in Benin, with serogroup distribution of Neisseria meningitidis

Chakir Ishola Bello, Cyriaque Comlan Degbey, Yves Eric Denon, Sessi Armel R. Hinvi, Lamine Baba-Moussa

BackgroundHealthcare workers in paediatric settings are in close contact with vulnerable children and may serve as reservoirs and vectors for the transmission of respiratory pathogens such as Streptococcus pneumoniae, Neisseria meningitidis, and Haemophilus influenzae. In the African meningitis belt, data on asymptomatic carriage of these pathogens among healthcare workers remain scarce. This study aimed to assess the nasopharyngeal carriage of these three major meningitis-associated bacteria among paediatric healthcare personnel in Benin.MethodsA cross-sectional study was conducted from September 2023 to December 2024 among 221 healthcare workers working in paediatric and neonatology units. Nasopharyngeal swabs were collected and analysed using quantitative real-time Polymerase Chain Reaction to detect S. pneumoniae (lytA), N. meningitidis (sodC), and H. influenzae (hpd3), as well as to characterise N. meningitidis serogroups and detect H. influenzae type b. Sociodemographic and clinical data were collected through a structured questionnaire. Associations between participant characteristics and pathogen carriage were assessed using chi-square tests and binary logistic regression, with a significance threshold set at p < 0.05.ResultsThe nasopharyngeal carriage prevalence was 24.4% (54/221) for S. pneumoniae, 11.8% (26/221) for H. influenzae, and 10.0% (22/221) for N. meningitidis. Among the meningococcal carriers, 8 (36.4%) carried serogroup Y, 3 (13.6%) serogroup X, and 1 (4.5%) serogroup A. The remaining positive samples did not belong to any of the major vaccine-targeted serogroups (A, B, C, W, X, Y). None of the Haemophilus influenzae-positive samples were of serotype b. Pneumococcal carriage was significantly associated with younger age (18–35 years), and residence within the meningitis belt (p < 0.05). Similarly, Neisseria meningitidis carriage showed a significant association with residence in the meningitis belt. In contrast, no significant association was o

Nasopharyngeal colonisation dynamics of bacterial pathogens in patients with fever in rural Burkina Faso: an observational study

Liesbeth Martens, Bérenger Kaboré, Annelies Post, Christa E. van der Gaast-de Jongh, Jeroen D. Langereis, Halidou Tinto, Jan Jacobs, André J. van der Ven, Quirijn de Mast, Marien I. de Jonge

BackgroundNasopharyngeal colonisation with clinically relevant bacterial pathogens is a risk factor for severe infections, such as pneumonia and bacteraemia. In this study, we investigated the determinants of nasopharyngeal carriage in febrile patients in rural Burkina Faso.MethodsFrom March 2016 to June 2017, we recruited 924 paediatric and adult patients presenting with fever, hypothermia or suspicion of severe infection to the Centre Medical avec Antenne Chirurgicale Saint Camille de Nanoro, Burkina Faso. We recorded a broad range of clinical data, collected nasopharyngeal swabs and tested them for the presence of Streptococcus pneumoniae, Haemophilus influenzae, Moraxella catarrhalis, Staphylococcus aureus and Klebsiella pneumoniae by quantitative polymerase chain reaction. Using logistic regression, we investigated the determinants of carriage and aimed to find correlations with clinical outcome.ResultsNasopharyngeal colonisation with S. pneumoniae, H. influenzae and M. catarrhalis was highly prevalent and strongly dependent on age and season. Females were less likely to be colonised with S. pneumoniae (OR 0.71, p = 0.022, 95% CI 0.53–0.95) and M. catarrhalis (OR 0.73, p = 0.044, 95% CI 0.54–0.99) than males. Colonisation rates were highest in the age groups < 1 year and 1–2 years of age and declined with increasing age. Colonisation also declined towards the end of the rainy season and rose again during the beginning of the dry season. K. pneumoniae prevalence was low and not significantly correlated with age or season. For S. pneumoniae and H. influenzae, we found a positive association between nasopharyngeal carriage and clinical pneumonia [OR 1.75, p = 0.008, 95% CI 1.16–2.63 (S. pneumoniae) and OR 1.90, p = 0.004, 95% CI 1.23–2.92 (H. influenzae)]. S. aureus carriage was correlated with mortality (OR 4.01, p < 0.001, 95% CI 2.06–7.83), independent of bacteraemia caused by this bacterium.ConclusionsAge, sex and season are important determinants of nasophary

National-scale simulation of human movement in a spatially coupled individual-based model of malaria in Burkina Faso

Robert J. Zupko, Tran Dang Nguyen, Amy Wesolowski, Jaline Gerardin, Maciej F. Boni

Malaria due to the Plasmodium falciparum parasite remains a threat to human health despite eradication efforts and the development of anti-malarial treatments, such as artemisinin combination therapies. Human movement and migration have been linked to the propagation of malaria on national scales, highlighting the need for the incorporation of human movement in modeling efforts. Spatially couped individual-based models have been used to study how anti-malarial resistance evolves and spreads in response to drug policy changes; however, as the spatial scale of the model increases, the challenges associated with modeling of movement also increase. In this paper we discuss the development, calibration, and validation of a movement model in the context of a national-scale, spatial, individual-based model used to study the evolution of drug resistance in the malaria parasite.

Native entomopathogenic Metarhizium spp. from Burkina Faso and their virulence against the malaria vector Anopheles coluzzii and non-target insects

Etienne Bilgo, Brian Lovett, Raymond J. St. Leger, Antoine Sanon, Roch K. Dabiré, Abdoulaye Diabaté

BackgroundGenetically enhanced Metarhizium pingshaense are being developed for malaria vector control in Burkina Faso. However, not much is known about the local prevalence and pathogenicity of this fungus, so we prospected mosquitoes and plant roots (a common habitat for Metarhizium spp.) for entomopathogenic fungi.ResultsOur investigations showed that Metarhizium spp. represented between 29–74% of fungi isolated from plant root rhizospheres in diverse collection sites. At low spore dosages (1 × 106 conidia/ml), two mosquito-derived M. pingshaense isolates (Met_S26 and Met_S10) showed greater virulence against Anopheles coluzzii (LT80 of ~7 days) than isolates tested in previous studies (LT80 of ~10 days). In addition, the local isolates did not cause disease in non-target insects (honeybees and cockroaches).ConclusionsOur work provides promising findings for isolating local Metarhizium strains for application in mosquito biological control and for future transgenic biocontrol strategies in Burkina Faso.Electronic supplementary materialThe online version of this article (10.1186/s13071-018-2796-6) contains supplementary material, which is available to authorized users.

Natural Interactions between S. haematobium and S. guineensis in the Republic of Benin

Hélène Moné, Stéphanie Minguez, Moudachirou Ibikounlé, Jean-François Allienne, Achille Massougbodji, Gabriel Mouahid

Schistosomiasis is a parasitic disease which affects millions of people around the world, particularly in Africa. In this continent, different species are able to interbreed, like Schistosoma haematobium and Schistosoma guineensis, two schistosome species infecting humans. The Republic of Benin is known to harbor S. haematobium, but its geographical situation in between Nigeria, Mali, and Burkina Faso, where S. guineensis was found, raises the question about the possible presence of S. haematobium/S. guineensis hybrids in this country. We conducted morphological analyses on schistosome eggs and molecular analyses on schistosome larvae (high resolution melting (HRM) analysis and gene sequencing) in order to detect any natural interaction between these two species of schistosomes. The morphological results showed the presence of three egg morphotypes (S. haematobium, S. guineensis, and intermediate). Three genotypes were detected by ITS2 rDNA HRM analysis: S. haematobium, S. guineensis, and hybrid, and their percentages confirmed the results of the morphological analysis. However, sequencing of the CO1 mtDNA gene showed that all the samples from Benin belonged to S. haematobium. Our results provide the first evidence of introgression of S. guineensis genes in S. haematobium in Benin.

Natural products from Zanthoxylum heitzii with potent activity against the malaria parasite

Christopher Dean Goodman, Ingvild Austarheim, Vanessa Mollard, Bertin Mikolo, Karl Egil Malterud, Geoffrey I. McFadden, Helle Wangensteen

BackgroundZanthoxylum heitzii (Rutaceae) (olon) is used in traditional medicine in Central and West Africa to treat malaria. To identify novel compounds with anti-parasitic activity and validate medicinal usage, extracts and compounds isolated from this tree were tested against the erythrocytic stages of the human malaria parasite Plasmodium falciparum and for inhibition of transmission in rodent malaria parasite Plasmodium berghei.ResultsHexane bark extract showed activity against P. falciparum (IC50 0.050 μg/ml), while leaf and seed extracts were inactive. Fractionation of the hexane bark extract led to the identification of three active constituents; dihydronitidine, pellitories and heitziquinone. Dihydronitidine was the most active compound with an IC50 value of 0.0089 µg/ml (25 nM). This compound was slow acting, requiring 50 % longer exposure time than standard anti-malarials to reach full efficacy. Heitziquinone and pellitorine were less potent, with IC50 values of 3.55 μg/ml and 1.96 µg/ml, but were fast-acting. Plasmodium berghei ookinete conversion was also inhibited by the hexane extract (IC50 1.75 µg/ml), dihydronitidine (0.59 µg/ml) and heitziquinone (6.2 µg/ml). Water extracts of Z. heitzii bark contain only low levels of dihydronitidine and show modest anti-parasitic activity.ConclusionsThree compounds with anti-parasitic activity were identified in Z. heitzii bark extract. The alkaloid dihydronitidine is the most effective of these, accounting for the bulk of activity in both erythrocytic and transmission-blocking assays. These compounds may present good leads for development of novel anti-malarials and add to the understanding of the chemical basis of the anti-parasitic activity in these classes of natural product.Electronic supplementary materialThe online version of this article (doi:10.1186/s12936-016-1533-x) contains supplementary material, which is available to authorized users.

Natural Resource Management Schemes as Entry Points for Integrated Landscape Approaches: Evidence from Ghana and Burkina Faso

Samson Foli, Mirjam A. F. Ros-Tonen, James Reed, Terry Sunderland

In recognition of the failures of sectoral approaches to overcome global challenges of biodiversity loss, climate change, food insecurity and poverty, scientific discourse on biodiversity conservation and sustainable development is shifting towards integrated landscape governance arrangements. Current landscape initiatives however very much depend on external actors and funding, raising the question of whether, and how, and under what conditions, locally embedded resource management schemes can serve as entry points for the implementation of integrated landscape approaches. This paper assesses the entry point potential for three established natural resource management schemes in West Africa that target landscape degradation with involvement of local communities: the Chantier d’Aménagement Forestier scheme encompassing forest management sites across Burkina Faso and the Modified Taungya System and community wildlife resource management initiatives in Ghana. Based on a review of the current literature, we analyze the extent to which design principles that define a landscape approach apply to these schemes. We found that the CREMA meets most of the desired criteria, but that its scale may be too limited to guarantee effective landscape governance, hence requiring upscaling. Conversely, the other two initiatives are strongly lacking in their design principles on fundamental components regarding integrated approaches, continual learning, and capacity building. Monitoring and evaluation bodies and participatory learning and negotiation platforms could enhance the schemes’ alignment with integrated landscape approaches.

Nature prescribing: emerging insights about reconciliation-based and culturally inclusive approaches from a tricultural community health centre

Anita Vaillancourt, Rebecca Barnstaple, Natalie Robitaille, Taylor Williams

AbstractThis commentary highlights the importance of social and nature prescribing programs reflecting culturally diverse perspectives and practices. Creating and holding space for Indigenous and other worldviews should be a key priority of nature prescribing, a relatively recent practice in Canada that recognizes and promotes health benefits associated with engaging in a variety of activities in natural settings. Central to designing and delivering nature prescribing that is culturally inclusive and grounded in fulfilling obligations of reconciliation is recognizing the ongoing dominance of Western worldviews and their associated implications for decolonizing and Indigenizing nature-based programming. Consciously working to expand Western values, with the aim of extending nature prescribing practices beyond mere nature exposure to fostering emotional connections to nature, is a critically important part of the ongoing development of nature-based interventions and nature prescribing.

Nature, Déterminisme et Origine des Mécanismes d'Isolement dans le Complexe Jaera albifrons (Isopodes, Asellotes)

    Michel Solignac
