Pharmacological Evaluation of the Bronchorelaxant Effect of Waltheria indica L. (Malvaceae) Extracts on Rat Trachea

Rainatou Boly, Zakaline Yabre, Mathieu Nitiema, Boubacar Yaro, Jules Yoda, Lazare Belemnaba, Sylvain Ilboudo, Noëla Hoho Estelle Youl, Innocent Pierre Guissou, Sylvin Ouedraogo

Waltheria indica L. (Malvaceae) is a plant used in Burkina Faso for the treatment of various ailments including asthma. The aim of the study was to evaluate the pharmacological relaxant effect of the leafy stem extracts of Waltheria indica and thereby verify claim of use in treating asthma. Aqueous decoction and hydroalcoholic extracts obtained from the powdered leafy stems were screened for the presence of some phytoconstituents. The in vitro relaxant effect of the two extracts was evaluated on acetylcholine- (ACh 10−5 M) and potassium chloride- (KCl 6 × 10−2 M) induced contractions on rat-isolated tracheal preparations. To examine whether the potassium (K+) channels are involved in the relaxant effect, glibenclamide, an ATP-sensitive potassium channel inhibitor, was used. Moreover, to assess the safety of the extracts, acute oral toxicity was carried out on mice. The phytochemical screening revealed the presence of alkaloids, flavonoids, saponins, steroids, triterpenoids, tannins, and coumarins in the hydroalcoholic extract. Tannins, steroids, triterpenoids, and coumarins were not detected in the aqueous decoction. With respective EC50 values of 1.517 ± 0.002 mg/mL and 1.433 ± 0.001 mg/mL on ACh-and KCl-provoked contractions, the hydroalcoholic extract was found more potent in relaxing the isolated rat tracheal preparations compared to the aqueous decoction. In the presence of glibenclamide, the relaxant effect of the hydroalcoholic extract (EC50 = 0.191 ± 0.002 mg/mL) increased and was higher than that of the aqueous decoction. At dose of 5000 mg/kg of body weight, the extracts did not produce deaths or any significant changes in the general behavior of mice. The results suggest that different mechanisms including modulation of calcium and potassium channels, particularly the ATP-sensitive K+ channels, could be involved in the relaxation effect. These findings could justify the traditional use of W. indica in the management of asthma.

SummaryObjectivesNeisseria meningitidis, together with the non-pathogenic Neisseria species (NPNs), are members of the complex microbiota of the human pharynx. This paper investigates the influence of NPNs on the epidemiology of meningococcal infection.MethodsNeisseria isolates were collected during 18 surveys conducted in six countries in the African meningitis belt between 2010 and 2012 and characterized at the rplF locus to determine species and at the variable region of the fetA antigen gene. Prevalence and risk factors for carriage were analyzed.ResultsA total of 4694 isolates of Neisseria were obtained from 46,034 pharyngeal swabs, a carriage prevalence of 10.2% (95% CI, 9.8–10.5). Five Neisseria species were identified, the most prevalent NPN being Neisseria lactamica. Six hundred and thirty-six combinations of rplF/fetA_VR alleles were identified, each defined as a Neisseria strain type. There was an inverse relationship between carriage of N. meningitidis and of NPNs by age group, gender and season, whereas carriage of both N. meningitidis and NPNs was negatively associated with a recent history of meningococcal vaccination.ConclusionVariations in the prevalence of NPNs by time, place and genetic type may contribute to the particular epidemiology of meningococcal disease in the African meningitis belt. Highlights•A prevalence of 10.2% of Neisseria infection was observed during the study.•Five Neisseria species were identified in nasopharyngeal samples.•High level of genetic diversity was observed in carried isolates.•Inverse relationship between carriage of Neisseria meningitidis and non-pathogenic Neisseria.

Phase 3 Evaluation of an Innovative Simple Molecular Test for the Diagnosis of Malaria and Follow-Up of Treatment Efficacy in Pregnant Women in Sub-Saharan Africa (Preg-Diagmal)

The malaria parasite Plasmodium falciparum (Pf) can sequester in the placenta resulting in low density of peripheral parasitemia and consequently in false negative malaria diagnosis (by microscopy) in pregnant women. Moreover, the use of rapid diagnostic tests (RDTs) in diagnostic strategies, including those for the detection of a malaria infection during pregnancy, is constrained by either persistent malaria antigen (histidine-rich protein 2; HRP2) after successful treatment, leading to false positive test results, or by false negative results as previously mentioned due to parasite sequestration (which is further exacerbated due to the low limited of detection [LoD] of conventional RDTs) or to HRP2 deletion. Recently, a direct blood polymerase chain reaction combined with a nucleic acid lateral flow immunoassay (dbPCR-NALFIA) has been developed, which circumvents these challenges and has demonstrated its diagnostic potential in phase 1 and 2 studies. The PREG-DIAGMAL trial presented in this manuscript will assess the diagnostic performance of dbPCR-NALFIA for the diagnostic of malaria in pregnant women and its potential to monitor treatment efficacy in these subjects. The work is ancillary embedded in an ongoing EDCTP funded trial, the PyraPreg project (PACTR202011812241529) in which the safety and efficacy of a newly registered Artemisinin-Based Combination (Pyronaridine-Artesunate) is being evaluated in pregnant women. This is a Phase 3 diagnostic evaluation conducted in 2 African countries: Democratic Republic of the Congo (DRC) and Burkina Faso. Pregnant women fulfilling the inclusion criteria of the PyraPreg study will be also invited to participate in the PREG-DIAGMAL study. Diagnostic accuracy will be assessed following the WHO/TDR guidelines for the evaluation of diagnostics and reported according to STARD principles. Due to the lack of a 100% specific and sensitive standard diagnostic test for malaria, the sensitivity and specificity of the new test will

Phenolic Content, Antioxidant Potential, and Antimicrobial Activity of Uvaria chamae (Annonaceae), a Food Plant from Burkina Faso

Kayaba Kaboré, Crépin Ibingou Dibala, Hemayoro Sama, Mamounata Diao, Marius K. Somda, Mamoudou H. Dicko

The study aimed to evaluate phenolic content and antioxidant and antibacterial potentials of the fractions of the hydroethanolic extract of Uvaria chamae leaves, a food plant from Burkina Faso. Thus, the hexane, dichloromethane, ethyl acetate, and butanol fractions of the hydroalcoholic extract after drying were used to determine phenolic compound content, antioxidant activity, and antimicrobial potential on strains of pathogenic bacteria responsible for food contamination. Phytochemical analyses were performed according to standardized methods, while antioxidant activity was evaluated by DPPH and FRAP methods. The antibacterial activity of the fractions was determined by diffusion and microdilution methods on the agar medium with gentamicin as a reference antibiotic. All the six strains, namely, Salmonella typhi ATCC 19430, Escherichia coli ATCC 8739, Staphylococcus aureus ATCC 25923, Pseudomonas aeruginosa ATCC 9027, Bacillus cereus ATCC 13061, and Listeria monocytogenes ATCC 7644, were sensitive to the fractions tested. Minimum inhibitory concentrations ranged from 37 µg·mL−1 to 1.67 mg·mL−1, respectively, gentamicin and butanolic fractions, while minimum bactericidal concentrations of the fractions ranged from 0.037 to 2.500 mg·mL−1 depending on the bacterial strain. Antioxidant activity varied significantly between fractions. For DPPH free radical scavenging activity, the butanol fraction was the most active, with an IC50 of 280 μg/mL, while the lowest activity (705 μg/mL) was recorded by the hexane fraction. Those of trolox and ascorbic acid used as standards were 80 and 100 μg/mL, respectively. Ferric reducing power (FRAP) ranged from 0.34 to 0.40 mmol EAA/g extract for the hexanic and ethyl acetate fractions, respectively. Phenolic compound contents also varied significantly between fractions. Butanoic and ethyl acetate presented the best contents of total phenolics and flavonoids, respectively. Significant and positive correlations were also recorded betwee

Phenology under bottom-up control: change in host quality induces diapause in parasitic wasps

Kévin Tougeron, Jacques Brodeur, Joan van Baaren, David Renault, Cécile Le Lann

When organisms coevolve, any change in one species can affect phenotypes and ecology of the other species. Upper trophic levels have to synchronize their life-cycle to both abiotic conditions and lower trophic level species phenotypic variations and phenology. The role such interactions play in ecosystems is central, but their mechanistic bases remain underexplored. We tested the effect of seasonal variation in host quality on parasitoid diapause induction by using viviparous and oviparous female morphs of the pea aphid Acyrthosiphon pisum. Aphidius ervi parasitoids from two populations of contrasted climatic origin (harsh vs. mild winter areas) were allowed to parasitize each morph in a split-brood design and were next reared under either fall-like or summer-like temperature-photoperiod conditions. We found that oviparous morphs, present before winter, are cues per se for diapause induction; parasitoids entered diapause at higher levels when developing in oviparous hosts (19.4 3.0%) than in viviparous ones (3.6 1.3%), under summer-like conditions (i.e., when oviparous aphids appear in the fields). This pattern was only observed in parasitoids from the harsh winter area, suggesting local adaptations to variation in host quality as an overwintering cue. Analyses of the relative proportion of forty-seven metabolites and lipid reserves in both aphid morphs produced under the same conditions suggest parasitoids response to be mainly influenced by chemical cues derived from the host, with higher proportion of polyols and sugars, and more fat reserves being found in oviparous morphs. Host physiological state thus varies across the seasons and represents one of the multiple environmental indicators of upcoming detrimental conditions, affecting parasitoid overwintering strategies and diapause. Our results underline coevolutionary processes between hosts and parasitoids in their area of origin, likely leading to phenological synchronization with the environment. We point out

Traduction

    Fabian Lo Monte, Caroline Gros

BackgroundThe conjugate vaccine against serogroup A Neisseria meningitidis (NmA), MenAfriVac, was first introduced in mass vaccination campaigns of the 1-29-year-olds in Burkina Faso in 2010. The aim of this study was to genetically characterize meningococcal isolates circulating in Burkina Faso before and up to 13 months after MenAfriVac mass vaccination.MethodsA total of 1,659 meningococcal carriage isolates were collected in a repeated cross-sectional carriage study of the 1-29-year-olds in three districts of Burkina Faso in 2010 and 2011, before and up to 13 months after mass vaccination. Forty-two invasive isolates were collected through the national surveillance in Burkina Faso in the same period. All the invasive isolates and 817 carriage isolates were characterized by serogroup, multilocus sequence typing and porA-fetA sequencing.ResultsSeven serogroup A isolates were identified, six in 2010, before vaccination (4 from carriers and 2 from patients), and one in 2011 from an unvaccinated patient; all were assigned to sequence type (ST)-2859 of the ST-5 clonal complex. No NmA carriage isolate and no ST-2859 isolate with another capsule were identified after vaccination. Serogroup X carriage and disease prevalence increased before vaccine introduction, due to the expansion of ST-181, which comprised 48.5% of all the characterized carriage isolates. The hypervirulent serogroup W ST-11 clone that was responsible for most of meningococcal disease in 2011 and 2012 was not observed in 2010; it appeared during the epidemic season of 2011, when it represented 40.6% of the serogroup W carriage isolates.ConclusionsSuccessive clonal waves of ST-181 and ST-11 may explain the changing epidemiology in Burkina Faso after the virtual disappearance of NmA disease and carriage. No ST-2859 strain of any serogroup was found after vaccination, suggesting that capsule switching of ST-2859 did not occur, at least not during the first 13 months after vaccination.

Phenotypic changes in natural populations of Anopheles gambiae s.l. at the onset of the long dry season in tropical savannahs of Burkina Faso, West Africa

Wadaka Mamai, Karine Mouline, David Renault, Kevin Hidalgo, Kounbobr Roch Dabiré, Frédéric Simard

In the tropical savannahs with long dry seasons, malaria mosquito populations virtually disappear after the drying up of breeding sites to reappear in large numbers at the onset of next rainy season. While aestivation and long-distance migration are proposed as key strategies enabling these vectors to persist through the dry-season, the physiological, biochemical, and morphological traits underpinning these mechanisms remain insufficiently explored, particularly under natural field conditions. This study explored seasonal changes in Anopheles coluzzii, An. gambiae, and An. arabiensis at the onset of the dry season in the harsh savannahs of Burkina Faso, West Africa. Late-instar immature specimens were collected from two ecologically distinct sites, one with permanent and the other with only temporary breeding habitats, during the rainy season and the transitional period into the dry season. Larvae were reared to adulthood under natural conditions and several traits were analysed including ovarian development, sub-cuticular fat body hypertrophy, body size, and energy reserves. Gonotrophic dissociation was significantly more frequent in An. coluzzii at the onset of the dry season, indicating a shift toward reproductive arrest. All three species exhibited increased body size and cuticular fat deposits during the transitional period, though with species-specific differences. Notably, only An. coluzzii showed significant increases in energy reserves (proteins, lipids, and carbohydrates) during the transition period. These adaptive responses differed between the study sites, suggesting the influence of breeding habitats. The findings highlight that species within the An. gambiae complex engage in distinct phenotypic trajectories at the onset of the dry season, suggesting divergent adaptations and trade-offs in energy acquisition and allocation to survive during the dry season.

Phenotypic Detection of Carbapenemase and AmpC-β-Lactamase Production among Extended Spectrum β-Lactamase (ESBL)-Producing Escherichia coli and Klebsiella spp. Isolated from Clinical Specimens

Introduction: Data on antimicrobial resistance (AMR) are sparse across numerous African countries, as microbiological analyses are not routinely conducted and surveillance data are not collected. Accordingly, clinical samples are not routinely tested for carbapenem-resistant bacteria and, therefore, the general understanding of their prevalence in the region remains limited. Methods: Between January 2020 and June 2022, we collected extended spectrum β-lactamase (ESBL)-producing Enterobacterales (ESBL-PE) isolates from five hospitals in Burkina Faso. After an initial culture on ESBL-selective media, the species were identified using API20E and isolates were tested against 13 antimicrobial agents using the disc diffusion method on Mueller–Hinton (MH) agar. ESBL production was confirmed via a double-disc synergy test. Production of carbapenemases and AmpC-β-lactamases and phenotypic co-resistance were determined. Results: Among the 473 ESBL-PE, 356 were ESBL-E. coli (ESBL-Ec) and 117 were Klebsiella spp. (ESBL-K). Of these isolates, 5.3% were carbapenemase and 5.3% were AmpC-β-lactamase-positive. Three types of carbapenemases were identified: 19 NDM, 3 OXA-48-like and 1 VIM. Two isolates produced both NDM and OXA-48-like carbapenemases. Carbapenemase producers were detected at all levels of healthcare. Co-resistance rates were up to 85% for aminoglycosides, 90% for sulfonamides, 95% for fluoroquinolones and 25% for chloramphenicol. Fosfomycin resistance was 6% for ESBL-Ec and 49% for ESBL-K (49%). Conclusions: Some of the ESBL-Ec and ESBL-K co-produced carbapenemases and/or AmpC-β-lactamases at all healthcare levels and in various sample types with high co-resistance rates to non-betalactams. Carbapenem resistance is no longer rare, calling for testing in routine diagnostics, a comprehensive resistance surveillance system and infection control within healthcare.

Phenotypic Detection of Extended-Spectrum β-Lactamase and Carbapenemase-Producing Enterobacteriaceae from Wastewater Treatment Plants in Ouagadougou, Burkina Faso

Background/Objectives: The overuse of antibiotics has led to the emergence of antibiotic-resistant bacteria, posing a global public health challenge. Among these, extended-spectrum β-lactamase and carbapenemase-producing Enterobacteriaceae (ESBL-E and CPE) are of particular concern due to their potential to spread resistance in various environments. Understanding the prevalence and spread of these bacteria in the influent and effluent of wastewater treatment plants is essential. Methods: This study examined ESBL-E and CPE in wastewater from three WWTPs in Ouagadougou, Burkina Faso, and was conducted between February and August 2024. Phenotypic detection of ESBL-E was performed on the isolates using the double-disk synergy test and the combination disk test, whereas the CPE detection employed the combination disk test and the modified Hodge test. Results: A total of 250 Enterobacteriaceae isolates were found, with Escherichia coli, Klebsiella spp., Enterobacter spp., and Buttiauxella spp. the most predominant taxa. Phenotypic analysis revealed a high prevalence of ESBL-E, particularly in influent samples, with rates ranging from 55 to 98% across the WWTPs. CPE detection showed varying prevalence, with higher proportions identified in effluent samples, ranging from 37 to 68%, depending on the plant. These findings highlight the critical role of WWTPs in the persistence and potential spread of antibiotic-resistant bacteria. Conclusions: This study underscores the urgent need for improved wastewater treatment technologies and comprehensive monitoring systems to reduce the dissemination of ESBL-E and CPE in the environment. Addressing these challenges is crucial for mitigating the public health risks associated with antimicrobial resistance.

Phenotypic effects of concomitant insensitive acetylcholinesterase (ace-1R) and knockdown resistance (kdrR) in Anopheles gambiae: a hindrance for insecticide resistance management for malaria vector control

Benoît S Assogba, Luc S Djogbénou, Jacques Saizonou, Pascal Milesi, Laurette Djossou, Innocent Djegbe, Welbeck A Oumbouke, Fabrice Chandre, Lamine Baba-Moussa, Mylene Weill, Michel Makoutodé

BackgroundMalaria is endemic in sub-Saharan Africa with considerable burden for human health. Major insecticide resistance mechanisms such as kdrR and ace-1Ralleles constitute a hindrance to malaria vector control programs. Anopheles gambiae bearing both kdr and ace-1 resistant alleles are increasingly recorded in wild populations. In order to maintain the efficacy of vector control strategies, the characterization of concomitant kdr and ace-1 resistance, and their pleiotropic effects on malaria vector phenotype on insecticide efficacy are important.MethodsLarval and adult bioassays were performed with different insecticide classes used in public health following WHO standard guidelines on four laboratory Anopheles gambiae strains, sharing the same genetic background but harboring distinct resistance status: KISUMU with no resistance allele; ACERKIS with ace-1R allele; KISKDR with kdrR allele and ACERKDRKIS with both resistance alleles’ ace-1R and kdrR.ResultsLarval bioassays indicate that the homozygote resistant strain harboring both alleles (ACERKDRKIS) displayed slightly but significantly higher resistance level to various insecticides like carbamates (bendiocarb, p < 0.001; propoxur, p = 0.02) and organophosphates (chlorpyriphos-methyl, p = 0.002; fenitrothion, p < 0.001) when compared to ACERKIS strain. However, no differences were recorded between ACERKDRKIS and KISKDR resistance level against permethrin (Pyrethroid, p = 0.7) and DDT (Organochlorine, p = 0.24). For adult bioassays, the percentages of mosquitoes knocked down were significantly lower for ACERKDRKIS than for KISKDR with permethrin (p = 0.003) but not with deltamethrin. The percentage of mortality from adult bioassays was similar between ACERKDRKIS and ACERKIS with carbamates and organophosphates, or between ACERKDRKIS and KISKDR with pyrethroid and DDT. Concerning acetylcholinesterase enzyme, ACERKDRKIS strain showed similarAChE1 activity than that of ACERKIS.ConclusionThe presence of both kdrR

Hicham Sbai, Yassin Essatarra, Abdelkrim Shimi, Amal Ankouz, Bachir Benjelloun, Abdelmalek Oussaden, Khalid Ait Taleb, Nabil Kanjaa
